After extensive preparation and reading, I am nearly at the point where experimentation will begin. I, like my classmates, am entirely new to the type of lab work going into this course, so even the most essential procedures are new territory for me. In order to help prepare for extensive work of a scope I’ve never before done, I have made my best efforts to be proficient in the material and methods that will be used for the western blotting. Hopefully, this will lead to experimentation soon after thanksgiving break.
One way in which I completed this task was through the large variety of protocols available from the manufacturers I am purchasing my supplies. I simply used the exact process outlined by the manufacturer of my antibody and I’m filling in the gaps with supplemental resources. One single protocols will not give me all the information I need to complete the specific experiment I’m working on. In order to fix this I have found areas in my current protocol where I am unsure of the exact procedure. I then added these to a larger list of protocols in order to have a good idea of the exact procedure. Any small details I may not be able to find in literature, I have managed to find through instructional videos, such as the one here. And yes, that is Pirates of the Caribbean music.
